Objective: To establish a convenient method of separation and cultivation of macrophage. Methods: Abdominal cavity of were douched using DMEM medium without serum, and the contain cells was drawn out and were cultured in DMEM medium with 10% fetal bovine serum. The cells were identified by morphological observation, endocellular enzyme and phagocytosis. Results: The obtained cells were highly purified macrophages with morphologic characteristics of the macrophage in organism, favourable phagocytosis, acid phosphatase (ACP) and α-acetic acid naphthol esterase(α-ANE) activity.
