Determination of Rutin and Quercertin in Flos Sophorae Buds by Capillary Zone Electrophoresis with Electrochemical Detection

A simultaneous determination of rutin and quercetin in Flos Sophorae buds by capillary zone electrophoresis with electrochemical detection (CE-ED) was reported. The effects of potential of working electrode, pH and concentration of the running buffer, separation voltage and injection time on the CE-ED determination were investigated and discussed. Rutin and quercetin could be well separated in a 100 mmol/L borate buffer (pH 9.0) within 10 min. A 300 (m diameter carbon disc electrode was used as the working electrode positioned carefully opposite the outlet of the capillary in a wall-jet configuration at potential of 0.90 V (vs. SCE). There are excellent linearity between peak current and concentration of analytes in the concentration range of 7.5×10 -7 ～1.0×10 -3 mol/L for rutin and 5.0×10 -7 ～1.0×10 -3 mol/L for quercetin with the detection limits (S/N=3) of 4.34×10 -7 and 2.25×10 -7 mol/L, respectively. Relative standard deviations (RSD) of the peak currents of rutin and quercetin were 2.56% (rutin) and 4.11% (quercetin) based on 7 determinations of same sample containing 5.0×10 -4 mol/L rutin and quercetin. Average recoveries of 97.80% and 96.84% were obtained based on five determinations.
