Characterization and sequence analysis of 16S rDNA of a new species of high efficient biohydrogen production anaerobe

The isolation and identification of high efficient biohydrogen production anaerobic bacteria are the foundation of fermented biohydrogen production process by anaerobic digesting organic wastewater. It could make guick identification of taxon for the new isolates by sequencing 16S rDNA and phylogenetic analysis. The 16S rDNA base of isolate Rennanqilyfl were 1517 bp. The accession number of GenBank was AY332397. The isolate Rennanqilyfl is Gram + , rod, 2 polar flagella, longer flagellum. The strain Rennanqilyfl has capsule around germ. Strictly anaerobic, no spore - forming bacteria. The generation between two times was 7. 8 h. The optimum temperature is 38℃. The optimum growth pH value is 4. 5. The amount of hydrogen - producing (YH2)in a unit is 1902. 8 ml /L medium. The maximum hydrogen - producing rate(QH2)is 12. 9 mmol/(g·h) . The closest 16S rDNA sequence homology of the isolate Rennanqilyfl come from 5 genus of Clostridium et al. GenBank. The closest identy of 16S rDNA with Clostridium celluosi is 91% , others were 89-90%. The isolate Rennanqilyfl is a new species and possibly member of a new genus. The temporary nomenclature of the genus is Biohydrogenbacterium Gen. Nov. Sp. Nov.
