Optimum conditions of solid-state fermentation for white-rot fungi and for Its degrading straw

The effects of five single culture factors on lignin biodegradation ability of white-rot fungi in state-solid medium were optimized through the orthogonal experiments. The results indicated that the optimal fermentation for white-rot fungi could be reached when white-rot fungi was cultured after 8 days, 0.8% white-rot fungi (weight ratio) was inoculated, temperature was kept at 37℃, water content of the solid substrate was maintained by 85% and 0.3% Tween 80 was added respectively. In addition, the structure of the untreated straw and the treated straw degraded under the optimal fermentation condition was analyzed through ultraviolet spectroscopy and infrared spectroscopy. It could be observed that long-bond hydrocarbons of giant molecule difficult to be biodegraded were degraded into short-bond hydrocarbons of small molecule, which could be easily biodegraded. Compared with the structure of untreated straw, the structure of the treated straw was greatly altered.
