Effects of Cooking Oil Fume Particulate on Oxidative Stress Parameters in Lung Type II-Like Epithelium Cells of Fetal Mice

Objective To investigate the cytotoxicity of cooking oil fume particulate(PM2.5),which formed when peanut oil was heated,to fetal mice lung type Ⅱ-like epithelium cells(AECⅡ).Methods The mice alveolar typeⅡ epithelial cells were isolated from 18 days old fetuses of ICR mice.The cells were cultured at a density of 1×106 cells/ml,AECⅡ cells were treated with PM2.5 at the doses of 0 μg/ml(the solvent control,DMEM),25,50,100 and 200 μg/ml for 12,24 and 48 h.AEC Ⅱ cell proliferations were detected with MTT assays.The cells cultured with a density of 5×105 cells/ml were treated with OFPO at the the doses of 0 μg/ml(the solvent control,DMEM),12.5,25 and 50 μg/ml for 12,24 and 48 h,the activity of SOD,and the concentration of MDA and GSH in supernatant were determined with reagent kit.Results The survival rate of AECⅡ and the activity of SOD were decreased,the concentration of MDA was increased with a dose-time dependent manner,while the activity of GSH was decreased with a dose-dependant manner.Conclusion Cooking oil fume particulate may decrease the viability of AEC Ⅱ cells and induce lipid peroxidation in the cells.
