Enantioseparation of D,L-Tryptophan with Pre-mixed Chiral Mobile Phase by High Performance Liquid Chromatography

A high performance liquid chromatography (HPLC) method using pre-mixed L-phenyl-Alanine-Cu(Ⅱ) and methanol as chiral ligand mobile phase was described in this paper. Effects of mobile phase input ways (single pump or double pump), volume ratio of chiral ligand agent to methanol((90∶10~)83∶17), flow rates of eluent ((1.2)~(0.8 mL/min)) and temperature of column (30~(50 °C)) on the effi-(ciency of) resolution of D,L-tryptophan by HPLC were investigated. The thermodynamical parameters of D,L-tryptophan resolution with mobile phase on C18 column of HPLC are obtained as follows: ΔH_D=(-22.48 kJ/mol);ΔS_D=(-0.056 5 kJ/(mol·K));ΔG~(298.15)_D=(-5.66 kJ/mol);ΔH_L=(-22.26 kJ/mol);ΔS_L=(-0.052 6 kJ/(mol·K));ΔG~(298.15)_L=(-6.58 kJ/mol).
