Determination of sucrose esters on leave surface of tobacco by TLC scanning

With 7 different flue-cured tobacco including Honghuadajinyuan,Yunyan85,K326,K346,NC89,NC82 and G80 taken as experimental materials,the sample was separated by using toluene-ethyl acetate-methanol-water(10∶5∶4.5∶0.2 in volume ratio)as a mobile phase.Sucrose esters were developed by ascending technique on the silica gel G plate,visualized by urea-phosphori,acid-n-butanol with blue spots.Singal wavelength TLC-Scanning(λ=610 nm)was used.The results showed that TCL-Scanning area is stable in 4 h.Sucrose esters develop blue spots at 75 ℃for 10 min.The linear range of sucrose ester is 80~400 μg,and the correlation coefficient is 0.992 9.The average recovery is 96.63% while RSD is less than 3.5%.
