Detection of Salmonella with Helicase-dependent isothermal DNA amplification

Aiming at establishing Helicase-dependent isothermal DNA amplification(HDA),and providing a simple and rapid method for the detection of Salmonella and other pathogens.A set of highly specific primers was synthesized to target the invA gene of Salmonella.Through optimizing the conditions,the HDA method for detection of Salmonella was established.Four strains of Salmonella and six other pathogens was used to detect the method’s specificity.Dilution method was used to assess its sensitivity.The result shows that the HDA could be completed in the temperature constant bathing pot,with 75 minutes of amplification.The method presented 100% of positive rate for Salmonella,without any false positive one;12 animal feeds samples were detected for samonellai,ts positive rate was determined to be 25%.So,HDA method is very suitable for primary laboratory.
