Determination of icariin in rat plasma in different time by improved RP-HPLC after oral administration of epimedium compound preparation

To built a sensitive,specific and stable RP-HPLC combined with simple and efficient preparation for determination of icariin in rat plasma after intragastric administration with a epimedium compound preparation.Using ethylether to deal with the plasma sample,separation of icarrin,other constituents of compound preparation,and possible endogenous compounds were accomplished on Shim-pack VP-ODS chromatographic column by gradient elution.The results showed that the standard curve of icarrin was linear in the range of 0.063 8～10.200 0 μg/mL.The method is accurate and precise,within-day precision and day to day precision are 5%,extraction recovery is 85%,the sample is stable in 24 h.The improved method can be used for analysis of icarrin in plasma after intragastric administration with compound recipe.
