Synthesis of Five Deuterium Substituted 2-pyrimidinyloxy-N-arylbenzylamines with Herbicidal activity

In order to prepare isotope internal standards in quantitation of the residues of five novel herbicides,the methoxyl-deuterated forms of five ingredients were synthesized from(2H4) methanol in six steps via nucleophilic substitution,oxidation,condensation,and reduction reactions,respectively. The deuterium substituted compounds are isopropyl 4-( 2-chloro-6-( 4,6-di(2H3) methoxypyrimidin-2-yloxy) benzylamino) benzoate,propyl 4-( 2-chloro-6-( 4,6-di(2H3) methoxypyrimidin-2-yloxy) benzylamino) benzoate,5-bromo-N-( 2-chloro-6-( 4,6-di(2H3) methoxypyrimidin-2-yloxy) benzyl) pyridin-2-amine,4-bromo-N-( 2-chloro-6-( 4,6-di(2H3) methoxypyrimidin-2-yloxy) benzyl) aniline,and2,5-dichloro-N-( 2-chloro-6-( 4,6-di(2H3) methoxypyrimidin-2-yloxy) benzyl) aniline. Their structures were elucidated by NMR,MS,FTIR,and UV,and their chemical purities are all great than 98%,determined by HPLC. These deuterium substituted compounds could be used as isotope internal standards in quantitation of herbicide residue in agricultural products and its downstream foodstuff by HPLC-MS / MS.
