Cloning of 16S rRNA sequence of the Dunaliella salina chloroplast

Aim: To clone and identify a 16S rRNA sequence of the Dunaliella salina chloroplast. Method: The 16S rRNA sequence of the chloroplast of Dunaliella salina was amplified using primers which were designed according to the evolutionally high conserved regions determined by analyzing the 16S rRNA sequences of the chloroplast of four green algae. Results: The cloned sequence of 1 110 bp had 85.0%, 79.9%, 81.3% and 81.6% identity with the 16S rRNA sequences of four green algae Chlamydomonas reinhardtii, Chlorella vulgaris, Nephroselmis olivacea and Mesostigma viride, respectively. Conclusion: The cloned sequence proves itself a 16S rRNA sequence of the Dunaliella salina chloroplast.
