Construction of transformation vector pDS16S-CAT for chloroplast of Dunaliella salina

Aim: To transfer chloramphenicol acetyltransferase (CAT) gene to the chloroplast of Dunaliella salina by homologous recombination. Method: Cloned 16S rRNA sequence of the chloroplast of Dunaliella salina was used as homologous fragment to construct a transformation vector pDC16r-CAT which was transferred to the cells of Dunaliella salina, using microprojectile delivery. Results: The transformed strain of Dunaliella salina cells survived in media containing chloramphenicol for 3~4 weeks and the transformants exhibited obvious chloramphenicol resistance, suggesting that the CAT gene be transferred to the chloroplast of Dunaliella salina.Conclusion: It is available to transform the chloroplast of Dunaliella salina and correct homologous sequences are necessary for stable transformation.
