Transcription activities of the atpA promoter of Chlamydomonas reinhartii chloroplast in Dunaliella salina

Aim: To prove the transcription activities of the atpA promoter of Chlamydomonas reinhartii chloroplast in Dunaliella salina. Method: The vector pSP71-atpA-EGFP was constructed by inserting the reporter gene ”enhanced green fluorescent protein”(EGFP) between the atpA promoter and the rbcL terminator of chloroplast of Chlamydomonas reinhartii, and transferred to the chloroplast of Dunaliella salina. The expression of EGFP controlled under the atpA promoter was observed through fluorescent microscope to prove the activities of the atpA promoter in Dunaliella salina. Results: The expression of EGFP was observed in the chloroplast of Dunaliella salina under fluorescent microscope. Conclusion: The atpA promoter of the chloroplast of Chlamydomonas reinhartii shows activities in Dunaliella salina, suggesting that it may be used to transform the chloroplast of Dunaliella salina.
