Experimental study on prevention of alcohol-induced osteonecrosis of the femoral head with siRNA adenovirus vector to suppress the expression of PPARγ mRNA

Objective:To observe the preventative effects on alcohol-induced osteonecrosis of femoral head(ONFH) with siRNA adenovirus vector targeting PPARγ gene to suppress adipogenic differentiation in marrow stromal cells(MSCs) of rabbit induced by alcohol.Methods:RNAi technique was used,recombinant adenovirus vector for expressing PPARγ siRNA was constructed;The vectors were infected to rabbit MSCs in vitro and the cells of femoral head in vivo,the expression levels of PPARγ mRNA,PPARγ protein,content of intracellular triglyceride,ALP activity in cells,and osteocalcin in culture media were determined;The number of adipocytes was counted under light microscope;The pathological changes of the femoral head were studied.Results:The expression levels of PPARγ mRNA,the number of adipocytes,content of triglyceride,ALP activity and osteocalcin in culture media in group siRNA approximated to the results in group Normal,there were statistically significant differences compared with the results in group Model,Control O and Control 1 (P0.05),there were no statistically significant differences compared with the results in group Normal (P0.05).In vivo,the expression level of PPARγ mRNA decrease,the percentage of empty osteocyte lacunae,average diameter of the max adipocyte,trabeculace area fraction in femoral heads of group siRNA approximated to the results of group Normal (P0.05).Conclusion:siRNA adenovirus vectors was able to inhibit expression of PPARγ mRNA and alcohol-induced adipogenesis of MSCs in vitro and the cells of femoral head in vivo,prevent onset of alcohol-induced ONFH.
