Improved Synthesis of 2,6-Dimethyl-3,5-diethoxycarbonyl-1,4-dihydropyridine Without Solvent and Its Aromatization

DING Ying-hong,ZHOU Zhun-liang(Guangdong College of Pharmacy,Guangdong Guangzhou 510224,China)  
2,6-Dimethyl-3,5-diethoxycarbonyl-1,4-dihydropyridine was synthesized by the reaction of formaldehyde,ethyl acetoacetate and ammonium acetate with 88.15% yield without solvent and under normal heating condition.The optimum conditions were: reaction temperature,57℃;and reaction time,30 min.2,6-Dimethyl-3,5-diethoxycarbonyl-1,4-dihydropyridine was aromatized with 97.20% yield;by ammonium persulfate in the presence of some little acetone and water.The aromatization reaction conditions were;reaction temperature 27℃;n(dihydropyridine)∶n([o])=1∶1;reaction time,3~4 min.
