Determination of γ-aminobutyric acid in Tartary buckwheat by UPLC-TOF/MS

OBJECTIVE To detect the content of γ-aminobutyric acid(GABA) in the Tartary buckwheat by UPLC-TOF/MS.METHOD By the reaction of GABA and the hydrochloric acid in butanol,the GABA easter was formed.Waters Acquity UPLC was used for the determination with BEH C18(100 mm×2.1 mm,1.7 μm)as the chromatographic column,flow phase acetonitrile-methanol-0.01 mol ·L-1 ammonium acetate(45:35:30),flow rate 0.2 mL·min-1,column temperature 30 ℃.Waters LCT Premier XE(TOF/MS),electronic spray ionization source(ESI) and the poison(mode V)was selected.RESULTS When the concentration of GABA within 0.4831-4.8312 μg,the average recovery was 97.28% with r of 0.9996.CONCLUSION This method was simple,precious and reproducible,thus could be used to determine the GABA in the tartary buckwheat.
