Study on selective marker for genetic engineering of Undaria pinnatifida gametophytes

The sensitivity of Undaria pinnatifida gametophytes to PPT(phosphiothricin) at different temperatures(10.0,15.0,20.0,25.0℃, ±0.5℃) was tested.The results of LD50 and 95% Confidence Range of PPT showed that U.pinnatifida gametophytes were sensitive to PPT;increasing the cultivation temperature could significantly decrease the LD50 of PPT.At the sixth day,the LD50 dropped from 15.78 mg/L at 10.0℃ to 11.56 mg/L at 20.0℃ and 10.33 mg/L at 25.0℃.The results mentioned above indicated that PPT could be a promising selective pressure for U.pinnatifida gametophytes transgenic research.
