THE EFFECT OF SEPIA ON INTRACELLULAR Ca~(2+) 、 ATP CON CENTRATION AND MITOCHONDRIAL Ca~(2+) /Ma~(2+) -ATPase ACTIV- ITIES IN H_(22) CANCER CELLS

The effects of Sepia on the intracellular Ca~(2+)、ATP concentrations and mitochondrial Ca~(2+)/Mg~(2+) -ATPase activities were studied in H_(22) cancer cells by using fluorescent probe and luciferin. The results showed that [ Ca~(2+) ] i decreased 2. 3 and 3. 7 times, mitochondrial Ca~(2+) /Mg~(2+) -ATPase activi ties diminished 28% and 58%, [ATP] i rose 77% and 83 %, respectively. The results suggested that Sepia probably diminished [ Ca~(2+) ] i, affected mitochondrial Ca~(2+) -dependent Ca~(2+)/Mg~(2+) -ATPase activi ties,thus reduced the amount of Ca~(2+) transported into mitochondria, lessened even eliminated the in hibitory effect of Ca~(2+) on ATP synthesis. This perhaps was one of the possible anti-cancer mechanisms of Sepia.
