Identification of the Protease-Producing Strain B1394 and Characterization of the Protease

The strain B1394 exhibiting the highest protease-producing activity was screened from 185 marine bacteria isolated from the marine sediments in South China Sea at a depth from 3 200—3 600 m.Its protease activity can reach 873 U/mL.B1394 was characterized and identified as Bacillus subtilis,based on the physiological and biochemical characteristics and analysis of 16S rDNA sequence.The optimal temperature and pH of the enzyme were found to be 60 ℃ and pH 8.0.The enzyme exhibited a good activity in low temperature and a good stability at 40—60℃.The enzyme could be stimulated by Mn2+,Mg2+and Ca2+.The enzyme could be inhibited by Hg2+,Fe3+,Cu2+, Zn2+and Fe2+.The enzyme activity was inhibited completely by PMSF.
