EXPERIMENTAL STUDIES ON THE HEMATOPOIETIC GROWTH FACTOR PRODUCTION INDUCED BY TOTAL SAPONINS OF PANAX GINSENG

Fibroblast,macrophage,lymphocyte and bone marrow stromal cell play a very important role in regulation of hematopoiesis.Saponins is known as one of the main effective fractions in Panax ginseng.By using the techniques of culture of hematopoietic progenitor cell in vitro and bio assay of interieukin 2, 6(IL 2, 6),the hematopoietic growth factor (HGF) production induced by total saponins of Panax ginseng(TSPG) was studied.The results indicated that the macrophage conditioned medium (MφCM),splenic cell conditioned medium(SCM),L cell conditioned medium(LcCM) and bone marrow stromal cell conditioned medium(BMSCM),prepared by treating these cells with TSPG(20mg/L) in vitro,had much stronger stimulating activities on the proliferation of BFU E,CFU GM,CFU MK;IL 6 activity in TSPG induced SCM,MφCM,and LcCM was markedly higher than that of control;IL 2 activity in TSPG induced SCM was higher than that of control,while TSPG induced MφCM,LcCM had no significant difference compared with that of control.This research suggested that TSPG may enhance the hematopoiesis by activating directly or indirectly on the hematopoietic stromal cells(macrophage,fibroblast)in hematopoietic inductive microenvironment,as well as lymphocyte to produce some HGF(such as GM CSF,Epo,BPA and IL 2, 6).This may be one of the mechanisms for the promoting vital energy and hematonic of panax ginseng.
