Distribntion and localization of sperm protein SP22 in human testis and sperm

Objective: To prepare rabbit antibody against human sperm protein SP22 by expressing recombinant htSP22 in a pro-karyotic system and study the localization of SP22 in human testis and sperm. Methods: Human testis gene SP22 was expressed in E. Coli via IPTG induction. The purified recombinant protein htSP22 was used to immunize rabbits. SP22 expression in human testis and sperm was detected by immunohistochemical and immunocytochemical staining with polyclonal antibodies. Results: The htSP22 protein was expressed successfully with a molecular weight of 22 000. The polyclonal antibody specifically recognized natural SP2'2 in human testis and sperm. Immunohistochemical and immunocytochemical staining showed strong signals from the spermatogenic cells resting in the most luminal region of the testicular tubuli and the surface of sperm head. Conclusion: The obtained polyclonal antibodies are specific to natural SP22, which only exists in spermatogenic cells resting in testicular tubuli and on the surface of human sperm head.
