Effect of obstructive sleep apnea hypoxia on autophagy of the hippocampal neurons after cerebral ischemia in rats

Objective:To investigate the effects of obstructive sleep apnea hypoxia(OSAH) on autophagy of the hippocampal neurons in rats with ischemia.Methods:Healthy male Wistar rats were randomly divided into sham operation group C SO group),ischemia/reperfusion group(I/R group),OSAH hypoxia for 7 days before ischemia/reperfusion group(IH7 + I/R group) and OSAH hypoxia for 21 days before ischemia/reperfusion group(IH21 + I/R group).Cerebral ischemia-reperfusion animals were prepared by improved Pulsinelli four vessel block(4-VO) for 24 hours.The morphological changes of hippocampal neurons were detected with H-E staining,and the expression of mTOR and beclin-1 in the hippocampus was detected by immunohistochemistry and RT-PCR Results:Compared with the SO group,the I/R group showed damages of hippocampal neurons;immunohistochemistry analysis showed that the number of mTOR and beclin-1 postive cells increased;RT-PCR analysis showed that the expressios of mTOR and beclin-1 were increased.Compared with the I/R group,OSAH hypoxia ischemia groups exacerbated the damage of hippocampal neurons;immunohistochemistry analysis showed that the number of mTOR and beclin-1 postive cells was increased;RT-PCR showed that the expressions of mTOR and beclin-1 were increased at IH7 +I/R group.The above changes were more apparent in the IH21 + I/R group.Conclusion:OSAH hypoxia exacerbates autophagic damages in the hippocampus after ischemia in rats.
