Acquirement of Insect-resistant Transgenic Plants of Brassica campestris ssp.Chinensis L.

The cotyledons of aseptic seedlings of Brassica campestris ssp chinensis L.cv.“Zhongjiaoheiye” and “Aijiaoheiye”were used as explants.The adventitious buds were obtained from these cotyledons on the modified MS medium supplemented with 2 mg/L CPPU,0.1 mg/L NAA and 7.5 mg/L AgNO 3.The inductive rates of adventitious buds were up to 32.52%,4.35% and 4.97%,11.55%,respectively,in cotyledon petioles or cotyledons of Zhongjiaoheiye and Aijiaoheiye.Through agrobacterium-mediated transformation,the recombinant plasmids carrying the CpTI gene and NPTⅡ gene were introduced into cotyledons of the two varieties.Regenerated plants with kanamycin resistance were obtained in the progeny (T 1),and the CpTI gene was also delivered to the progeny(T 1).The insect-resistance of the transgenic plants was maintained in the progeny(T 1).
