Study on preparation of complex amino acids by hydrolyzing sesame protein using co-immobilized double enzymes

In this article,in order to study the condition and stability of coimmobilized double enzymes as well as its application in hydrolyzing sesame residum protein,the authors tried to make use of Sodium acetylide as carrier and glutaraldehyde as cross-linking agent for the co-immobilized double enzymes.And the result showed that the optimal conditions of coimmobilization of double enzymes were: 3 g·dL-1 of Sodium acetylide concentration,25 g·L-1 of glutaraldehyde,0.2 g·dL-1 of CaCl2,and the recovery activity of coimmobilized enzyme was 51.28%.In addtition,the optimal conditions of hydrolyzing sesame protein were 1∶25 ratios of solid to liquid(w/v),pH 6.0,60 centigrade temperature,seven hours,added 5 g·dL-1 of co-immobilized double enzymes.The highest amount of amino nitrogen was 20.65 mg·g-1.The activity of co-immobilized double enzymes remained more than 50% after repeated 6 times.The amount of 15 kind amino acids was wholeness in sesame protein.
