Protective effect of acupuncture in acupoints of Governor Vessel on dopaminergic neurons protection and influence on ultrastructure in mice with Parkinson's disease

Objective To investigate the curative effect and mechanism of acupuncture in acupoints of Governor Vessel for treating Parkinson's disease (PD).Methods All male C57BL/6 mice were randomly divided into 5 groups,including the normal group,model group,madopar group,acupuncture group and acupuncture combining with madopar group (acu+mad group).The model was established and mice survived for 28 days.The mice motor coordination was measured by using pole test.The content of dopamine and its metabolic products in forebrain were detected by applying electrochemical HPLC internal standard method.The positive neurons of tyrosine hydroxylase in the substantia nigra of midbrain were detected by using immunohistochemistry.The ultrastructural changes of neurons in the substantia nigra were observed with transmission electron microscope (TEM).Results The pole-climbing time were shorter significantly in 3 treatment groups than that in the model group (P0.05),and that was much shorter in the acu+mad group than that in the acupuncture group and madopar group (P0.05).The content of dopamine and its metabolic products in forebrain increased significantly in the acu+mad group than those in the acupuncture group (P0.05),and the positive neurons of tyrosine hydroxylase in substantia nigra were higher than those in the madopar group (P0.05).The TEM result showed that the degree of mitochondria injury in the neurons of substantia nigra was lower in the acupuncture group and acu+mad group than that in the model group and madopar group.Conclusion Acupuncture in acupoints of Governor Vessel combining with madopar can relieve the abnormal behaviors in PD mice.The mechanism may be related to that the decrease of loss of dopaminergic neurons in the substantia nigra,and the relief of mitochondria injury.
