The Expression and Significance of Tissue Transglutaminase in Human Chronic Kidney Disease

Objective To investigate the expression of tissue Transglutaminase(tTG)in renal tissue of patients with chronic renal diseases,so as to analyze the clinical significance of tTG.Methods Biopsy samples from 37 patients with chronic renal disease were divided into four groups according to their primary disease,including LN group(n=15),PSGN group(n=7),IgA N group(n=9)and Nephritis of anaphylactoid puerperal group(n=6).Meanwhile,control group was established with 5 normal renal tissues.Expression of tTG in renal tissues were analysed by immunohistochemical staining.Relationship between the tTG in renal tissues and clinical data were analysed.Results Expression of tTG in diseased renal tissues was significantly higher than that in normal group(P0.05),but no significant difference was observed in each diseased group.Expression of tTG in renal glomerulus and renal tubule were higher in LN Ⅱgroup than in LN Ⅳgroup.In LNgroup,the expression of tTG in renal tissue was in positive correlation with pathological change of the glomerulus,but in negative correlation with serum C3 and C4.Conclusions tTG was overexpressed in the renal tissues of human chronic renal disease,but its expression is independent of the original etiology.The severer renal injured,the more tTG expressed.tTg may therefore play an important role in the pathogenesis of renal fibrosis in patients with CKD and can be considered a potential therapeutic target.
