Experimental Study of the osteogenesis of cultured osteoblast derived from the rabbit marrow in vivo

Objective:To establish a new cultured cell model of rabbit osteoblasts in vitro and study it's biological characteristic,to observe it's osteogenesis in vivo.Methods:Derived from the rabbit stromal cells of bone marˉrowfrom the trochanteric regoin jof femur of rabbit and cultured the cells with special cultural fluid in vitro.In abouttwoweeks cells lined up inone layer.Thencells were generated and theirbiological characteristic was examˉined by phase_contrast microscope,scanning electron microscope,transmission electron microscope,H_E stain,ALP stain,typeⅠcollagen stain and caluaria stain.The cells_ePTFe compound were cultured for one week and planted into the rabbit muscle.The planted materials were obtained at4,8weeks after operation.The specimens were assessed by histology method in order to assess the bone_forming ability of the cultured cells.Results:Morˉphologically,the cultured cells grewwell in primaryculture.Theylined up in one layer and appeared as flat,mulˉti_morphological cells withlongorshortprotrusions.Furthermore,theircellularoutline had a fibroblast_like appearˉance.Then the cells continued to crowd,and multilayer regions were formed.Transmission electron microscope examinationthe cytoplasmis richinribosome,endoplasmic reticulumand Golgi bodies,demonstratedwell symthetˉic and secretive fundion in accord with the characteristics of osteoblast.The biochemistry and histochemicalstainsshowedthatculturedcellswererichinALP,TypeⅠcollagenandthecalcificationinvitrowhichwere oftenusedasabiochemicalandhistochemicalmarkerfortheiridentificationandfortheevaluationofosteogene sis.AftercompoundedwithePTFe,cellsgrewwell.At4weeksafteroperation,therewerestillmanyosteoblast aroundthenewly_formedbonematrix.At8weeksafteroperation,cellsreducedandthenewly_formedbone lookedliketypicalbonetissue.Conclusion:Theculturedcellsderivedfromtherabbitbonemarrowstromal wererabbitosteoblasts.Thiscouldbeanewmodleofculturedosteoblastinvitro.
