The study of altered succinate dehydrogenase gene expression by Gypenosides during rat tongue OLK carcinogenesis induced by 4-Nitroquinoline 1-Oxide.

Objective:To examine the inhibitory effect of Gypenosides on 4-nitroquinoline 1-oxide(4NQO)-induced tongue carcinogenesis in rats and the dynamic effect of GP on SDH gene expression.Method:A total of 70 Wistar rats were divided into four group s:model group(n=25),administration group(n=30),normal control group(n=5) and herb control group(n=10).4NQO dissolved in drinking water was administered orally to induce tongue carcinogenesis in rats.Gypenosides was injected into the stomach of the rats during oral carcinogenesis.Rats were sacrificed at 9,13,20,24 and 32 weeks from the beginning of the experiment.The tongues of the rats were dissected for gross and histological assessment,and Real-time quantitative PCR was employed to detect the gene expression of SDH in the specimens.Result:Compared with the model group,the incidence of dysplasia were suppressed by Gypenosides(P 0.05).the expression level of SDH gene was obviously elevated in administration group(P 0.01),especially in 20 and 24 weeks(P 0.05).Conclusion:Administration of Gypenosides effectively suppressed 4NQO induced tongue carcinogenesis.Gypenosides may exert its chemopreventive effects by up-regualted expression of SDH to restore Tricarboxylic acid cycle.
