Role of microRNA-23a in pathogenesis of pulmonary hypertension in rats

Objective To explore the role of microRNA(miR)-23a in pathogenesis of pulmonary hypertension(PH)in rats.Methods Twenty-four male rats were randomly divided into PH group(n=12)and control group(n=12).Rat model of PH was established by hypoxia/high carbon dioxide method.Expression level of miR-23a in pulmonary arteria,content of tumor necrosis factor(TNF)-a in bronchiole alveolar lavage fluid(BALF)and apoptosis of endothelial cells in pulmonary small arteria in rats of two groups were detected by real-time quantitative polymerase chain reaction(RT-qPCR),enzyme-linked immunosorbent assay(ELISA)and terminal deoxynucleotide transferase-mediated d-UTP notch terminal labeling technique(TUNEL)respectively,and then to compare their results.Results Expression level of miR-23a in pulmonary arteria in PH group was(2.96 ±0.72)times of that in control group and the difference was statistically significant(P0.05).Content of TNF-a in BALF in PH group[(2 016 ±244)pg/ml]was significantly higher than that in control group[(1 228 ±118)pg/ml,P0.05].Compared with that in control group,apoptosis of endothelial cells in pulmonary small arteria increased obviously in rats of PH group.Conclusion MiR-23a may be related with inflammatory response caused by PH in rats,the up-regulation of which may be involved in the pathological process of PH in rats.
