Influence of Paraquat on Vesicular Monoamine Transporter 2 in Nigra-striatum of Mice

Aim:To explore whether the vesicular monoamine transporter 2 (VMAT2) was involved in pathogenic mechanism of parkinsonian mice induced by paraquat. Methods: Apply immuno-histochemistry and in situ hybridization techniques respectively to observe the changes of VMAT2 in striatum and the expression of VMAT2 mRNA in substantia nigra of parkinsonian mice induced by parquat taken orally. Results: Mice treated by paraquat (10 mg·kg -1)taken orally for two months displayed a marked hypoactive behavior.Levels of VMAT2 in striatum of mice were decreased by 42%(P0.01),and the expression of VMAT2 mRNA were also decreased by 37.2% compared to the mice treated by saline taken orally (P0.01).Conclusion:Paraquat could induce mice to appear a series of the behavior that were similar to Parkinson's disease.VMAT2 could be involved in the pathogenic mechanism of Parkinson's disease,because of the decreased levels of both the protein and mRNA expression.
