Synergistic Toxic Effect of Paraquat and Lactacystin on Substantia Nigra

Aim: To evaluate the synergistic toxic effect of paraquat and lactacystin on substantia nigra. Methods: 56 SD rats was randomly divided into four groups ( n = 14):NS group, lactacystin group (calling lact group for short), paraquat group, double factors group. animals of paraquat group and double factors group were fed with paraquat (0.5 mg · kg -1 ) for 8 weeks. Animals of NS group and lact group were fed with sodium saline as control. Then, animals of lact group and double factors group were injected with 2 μg lactacystin by sterotaxic nilateral injection into the substantia nigral pars compacta; animals of paraquat group and NS group were injected with sodium saline as control; the behaviors, TH positive cells and expression of alpha-synuclein of rats were observed. Results: Only animals of double factors group developed progressively bradykinetic, less activity, abnormal posture and displayed apomorphine-induced contralateral behavior; TH positive cells were significantly reduced compared to NS group; the numerical value reduced by double factors exceed the sum total reduced by lactacystin and paraquat ( P 0.05) . The expression of alpha-synuclein in double factors group was significantly increased compared to NS group and most of nigral cells in double factors group showed Lewy body. Conclusion: There is synergistic toxic effect between lactacystin and paraquat and the chronic exposure to paraquat can increase the risk for PD of rats with lightly impaired proteasome.
