Pharmacokinetics of rats after administrating harmine hydrochloride by injection of vein and intragastric administration

AIM: To study the pharmacokinetics and bioavailability of harmine hydrochloride capsule in rats. METHODS:An 8 h blood collection was done by carotid artery cannulation after rat administrating harmine hydrochloride at a dose of 10 mg·kg -1 by injection of vein and at a dose of 40 mg·kg -1 by intragastric administration, respectively. The plasma concentrations of harmine hydrochloride were measured by reversed phase high performance liquid chromatography (RP-HPLC). RESULTS: Plasma peak level reached at about 0.23 h with an average value of 2.958 μg·mL -1 after intragastrical administration. Average elimination half-life was 5.33 h. Bioavailability of harmine hydrochloride capsule was 19.65%. CONCLUSION: Harmine hydrochloride is absorbed rapidly by rats but with a lower bioavailability.
