Activating rules of astrocytes and microglias in mice after cerebral ischemia

Objective To compare the activating rules of astrocytes and microglias in mice after cerebral ischemia.Methods A mouse focal cerebral ischemic injury model of middle cerebral artery occlusion/reperfusion was established by modified Longa occlusion.Forty-eight Kunming mice were randomly divided into sham operation group(n=24),cerebral ischemia group(n=24).The mice in cerebral ischemia group were further divided into 1-,3-,7-and 14-day reperfusion groups 3h after ischemia(6in each group).Lesions in brain tissue were observed with HE staining.Expressions of GFAP and Iba1 were detected with immunohistochemical staining.Results The number of GFAP-positive products in cortical ischemic boundary zone was significantly larger in cerebral ischemia group on days 3,7and 14 than on day 1after reperfusion(P0.01).Iba1-positive microglias in cortical ischemic boundary zone were detected in cerebral ischemia group on day1 after reperfusion.The number of activated microglias increased significantly and manifested as"bush-like"cells on days 3and 7after reperfusion and as"ameboid"cells on day 14 after reperfusion.Conclusion Both astrocytes and microglias are activated in cerebral ischemia reperfusion.However,microglias are activated earlier than astrocytes.Astrocytes are activated only in ischemic boundary zone while microglias can migrate from ischemic boundary zone to ischemic central zone.
