Expression of macrophage migration inhibitory factor, MMP-2 and MMP-9 in nasopharyngeal carcinoma

Purpose To detect the expression of macrophage migration inhibitory factor (MIF), MMP-2 and MMP-9 in nasopharyngeal carcinoma (NPC), to correlate their expression level with patients' clinicalopathologic features, and to investigate the mechanism of invasion and metastasis of NPC cells. Methods 45 NPC biopsies were collected from the patients with NPC during the period of 1999~2003. Immunohistochemical staining was used for detecting the expression of MIF, MMP-2 and MMP-9 in NPC tissues, and the associations of these proteins with pathological and clinical features were statistically analyzed. Results In 45 cases, positive expression rate of MIF, MMP-2 and MMP-9 was 77.8%(35/45), 64.4%(29/45) and 71.1%(32/45) respectively. The expression of MIF and MMP-9 showed a correlation with lymph node metastasis. The higher expression of MIF or MMP-9 were always found in NPC neoplastic cells with cervical lymph node metastasis than that in NPC without lymph node metastasis (both P value were less than 0.05). Meanwhile, the mean percentage of MMP-9 expression in MIF positive group (50.2%±33.5%) was significantly higher than that (11.7%±22.7%) in MIF negative group (P0.01). The expression of MIF significantly correlated with the expression of MMP-9 in NPC (r s=0.492, P0.01). However, there was no relationship found in the expression of MMP-2 either with the expression of MMP-9 and MIF or with lymph node metastasis. In addition, the expression of MIF in NPC neoplastic cells with Schmincke growth pattern (67.4%±35.2%) was higher than that in NPC with Regaud growth pattern (32.9%±29.7%), there existed a significant difference in MIF expression of NPC between two growth patterns (P0.01). Conclusions The over-expression of MIF and MMP-9 in the neoplastic cells contributes to the metastatic potential of NPC, and MIF may be an important factor to influence the growth pattern of NPC.
