Simultaneous Determination of Tetracycline, Oxytetracycline,Chlorotetracycline, and Chloramphenicol in Cow Milk by RP-HPLC

A RP-HPLC method for simultaneous determination of tetracycline(TC),oxytetracycline(OTC),chlorotetracycline(CTC),chloramphenicol(CP) is described.The separation is based on a Spherisorb C18 column with acetonitrile-0.2g/L nitrilotriacetic acid(pH2.5)(V/V=20:80) as mobile phase and UV detection at 267nm.Effects of mobile phase composition and pH on the separation are investigated.In the chosen conditions,good linearity between peak area and the antibiotic concentration is observed,and the detection limit for the 4 antibiotics are OTC 0.35 mg/L,TC 0.30 mg/L,CTC 0.80 mg/L and CP 0.70 mg/L,respectively.The spike recoveries of the 4 antibiotics are OTC 105%,TC 90%,CTC 82% and CP 87%,respectively.
