Composition Analysis of High Molecular Weight Glutenin Subunits (HMW-GS) in Synthetic Hexaploid Wheats

One hundred and three synthetic hexaploid wheats (2n = 6x = 42, AABBDD) were characterized for the composition of high molecular weight glutenin subunits (HMW-GS) by using sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). A large of allelic variations at three complex loci (Glu-A1 ,Glu-B1 and Glu-D1) were observed. There were five allelic variations at the Glu-A1 locus, 12 allelic variations at the Glu-B1 and Glu-D1 loci, respectively. Additionally, at Glu-D1 locus, a few accessions expressed only y-type glutenin subunits with "null" subunit 1Dx. Meanwhile, an unknown novel subunit and some new HMW-GS compositions were found. These synthetic hexaploid wheats with plentiful HMW-GS variations may be useful for genetic improvement of common wheat.
