Study on the effect of several growth factors and DMEM on grafted fat survival

Objective We research the effects of DMEM,bFGF,IGF-1 and VEGF on the grafted fat survival to offer reference to clinical practice. Methods We divided fat harvested under same condition into 5 groups and infiltrated the 5groups of fat respectively in saline, DMEM, saline containing 20ng/ml bFGF, saline containing 20ng/ml IGF-1, saline containing 20ng/ml VEGF for one hour. And then we transplanted 5 groups of fat subcutaneously on the back of 5 groups of mice, 5 mice per group, injecting 5 points per mouse, 0.2ml per point. After 15 weeks, we dissected fat grafts, measured their volume and weight retention and put them in histopathologic examination. We compared volume and weight retention of each experimental group with that of control group. Results The volume and weight retention of the group DMEM was significantly more than that of control group(P0.05). The volume and weight retention of the other experimental groups were all same as that of control group(P0.05). The histopathologic characteristics of all grafts were same. Conclusion DMEM can improve the volume and weight maintenance of graft. bFGF,IGF-1 and VEGF didn′t display the effect of improving fat survival in our experiment condition.
