Using SRBC as Target to Evaluaet Spontaneous Cytotoxicity of PBMC

convenient metnoa to assay spontaneous cytotozicity ot numan peripnerai blood mononuclear cells (PBMC) was reported here. Using sheep erythrocyte (SRBC) as target, the SRBC, when cocultured with PBMC, was cytolyzed and the released Hb then catalyzed H2O2 to oxidize o-pheno-diamine. Thus the spontaneous cytotoxicity of PBMC to SRBC can be evaluated by colorimetry. The main experiment conditions, adequate amount of SRBC, ratio of effector to target, optimal coculturing time and temperature, were studied in this report.
