Purification and Properties of Cellulose Activity from A.usamii

The cellulose was extracted from solid state leavening of A usamii. The crude product was purified by (NH 4) 2SO 4 and chromatographed by Sephadex G-200 column gel. The CMC component of cellulose is 6 times of raw products. The study on the enzymology of CMC enzyme shows that the best reaction temperature is 60 ℃, pH is 4.0, and enzyme is relatively stable at 30~60 ℃. The 90 % activity can be held in pH is 3.0~5.0 for 30 min at 50 ℃. There are IR spectrum absorption bands of O-H,N-H,C-H,CO. The values of CMC (K m ,v max ) were 0.10 g/m and 0.40 mg/(ml·min) respectively.
