PROTECTIVE EFFECTS OF ANISODAMINE 0N LEFT VENTRICULAR DIASTOLIC FUNCTION DURING MYOCARDIAL ISCHEMIA IN RATS
Effects of anisodamine (654-2) during acute myocardial ischemial were investigated in20 anesthetized open-chest rats, The animals were randomly divided into 2 groups.Normal saline iv 0.5 ml/kg (control group) and 654-2 iv 5mg/kg (treated group) weregiven 15 min before coronary occlusion. In n(?)n-ligated rats all hemodynamic parameterswere decreased significantly after iv 654-2. No change occluded in the control. It wasshown that 654-2 possessed the effects of inhibiting heart vessels. The occlusion of LADcaused a marked change in -dp/dt_(max), LVEDP and T (time constant of isovolumic pressure fallsubsequent to -dp/dt_(max) 20 min after ligation in the control, -dp/dt_(max) decreased from 525±118to 336±114 kPa, LVEDP increased from -0.35±0.24 to 0.31±0.48 kPa and T increased from8.1±1.9 to 13.0±2.5ms, respectively. But changes were slighter less 525±132 kPa, LVEDPincreased from -0.37 0.33 to -0.16±0.11 kPa and T increased from 4.2±1.7 to 4.5±0.9ms.The degree of reduction on other parameters in treated group was less than that of thecontrol. The results suggest that anisodamine might have exerted protective effect on leftventricular diastolic function during myocardial ischemia.
