Effects of Total Flavonoids of Epimedium on Vascular Function of Diabetic Mice

OBJECTIVE To study the effects of total flavonoids of epimedium(TFE) on systolic and diastolic functions of thoracic aortic in diabetic mice and its possible mechanism.METHODS Kunming male mice were administered intraperitoneally with alloxan solution to induce type I diabetes and the control mice were injected with the same volume of saline.The diabetic mice were randomly divided into model group,low-dose TFE group,and high-dose TFE group according to the blood glucose.Serum fasting blood glucose(FBG),superoxide dismutase(SOD) and malondialdehyde(MDA) were tested 2 weeks after continuous administration.The vasoconstriction response to phenylephrine(Phe) and the vasodilatation response to acetylcholine(ACh),sodium nitroprusside(SNP) of isolated mice thoracic aorta were observed,also the vasodilatation effect of the TFE in vitro were observed in the normal group and model group.RESULTS After administrating TFE for two weeks,the level of FBG and MDA decreased,the level of SOD increased,and the endothelium-dependent relaxation of thoracic aortas significantly improved in the model group.CONCLUSION The TFE may have a protective effect on the vascular endothelial injury in diabetic mice through decreasing blood glucose and the anti-oxidative stress.
