Study on androgen receptor in the target tissues of normal men with the methods of Scatchard, Woolf and double reciprocal plots

In male, genital skins were the androgen target tissues, which were bountiful of androgen receptor (AR) in cytosol and nucleochylema. Eleven foreskin specimen were taken from the volunteers (2O～34ys) at the time of circumcision. Six rat livers were required just before the receptor assay, as positive and qualitative control. About 1. 0g tissures were homogenized directly, without tissue and cell culture, and cetrifuged to get cytosol. The nucleochylema were extracted with 0. 6 mol/L KCl buffer from the nuclear pellets. The cytosolAR and nuclear AR radioassay were performed with the specific ligand R1881 and 3 methods for analysing data of Scatchard, Woolf and double reciprocal plots. The cytosol AR was characterized by Bm 24. 03± 7. 19 pmol/g protein and Kd (15.13±7. 06)×10-10 mol/L, and nuclear AR, Bm 204. 37±119. 62 pmol/g protein, Kd (6. 38±4.14) ×10-10 mol/L. The resultsof Scatchard, Woolf and double reciprocal plots were identical on statistic (P0. 05). So the androgen receptor radioassay in the principle of Scatchard pl0ts was stable, reliable and practical.
