DETERMINATION OF AMINO ACID CONTENT IN FEEDSTUFFS WITH THE HELP OF HPLC

Using Lichro CART Superspher CH-8,4um,250×4mm i.d.as stable phase and acetonitrile-Na-Acetat as mobile phase,the amino acids in the standard solution and in the hydrolyzed feedstuff solutions were derivatized with 9-fluorenylmethyl chloroformate(FMOC) at room temperature,separated with the reversed-phase system by adjusting the elution gradient and polarity of the mobile phase,and finally determined by fluoresence detector at emmition 310 nm and excitation 260 am.The results showed that FMOC had high sensitivi- ty and stability.A linearity was obtained between the concentration of amino acid and intensity of fluoresence within the range of concentrations in our experiment.The CV%of amino acids determined from the standard solutions was less than 4%,with the exception of lysine(CV=9.6%,n=12).The CV of the different hydrolyzed feedstuff solutions was less than 10%.
