Advance in Molecular Biology of Plant Parasitic Nematodes and the Strategies of Engineered Resistance to Nematodes

Plant-parasitic nematodes cause severe damage to world apriculture. Current controls are inadequate and heavily dependent on hazardous nematicides. Molecular biology offers a most economically and environmentally preferred solutions and molecular interaction between plants and nematodes provide new ways for engineered resistance against plant-parasitic nematodes. The ability of sedentary nematodes to induce specialized feeding sites in plant roots is one of the most fascinating aspects of host-parasite interaction. Many genes become up-regulated, while some constitutive promoters are down-regulated in the feeding sites within days after infection. Generally, there are three options available for plant genetic engineering against nematodes: 1) cloning and expression of resisboce gene, 2) synthesis of anti-nematode proteins, such as collagenase enzymes, antibodies fragments and proteinases inhibitors, 3) the targeted destruction of nematodes feeding sites. Nematode-induced promoters are very useful for anti-nematode proteins expression at feeding sites. Alternatively, interfering with feed-cell development can be achieved by the localized expression of a cytotoxin such as barnase or an antisense gene, which, however, calls for a highly specific non-leaky promoter, which is active only in the feeding cells.
