A RAPD Marker Linked to the Resistant Gene to Powdery Mildew in Wheat Variety - Fu Zhuang 30

Powdery mildew (Blumeria graminis CD f. sp. tritici) is an important fungal disease in wheat. The most effective method to control the disease is to develop host resistance.Mapping Pm genes permits marker assisted selection in wheat improvement programs. A recessive Pm gene of a wheat variety-Fu Zhuang 30 confers a broad spectrum resistance to powdery mildew in China. Here, we reported a RAPD marker linked to this gene. A 628 bp fragment was specifically amplified with RAPD primer UBC405 (CTCTCGTGCG) in Fu Zhuang 30, but not in susceptible cultivars Nong Da 015 and Jing Hua No. 1. With bulk segregant analysis of 101 F2 progenies of Nong Da 015/ Fu Zhuang 30 and 62 F2 progenies of Jing Hua No. 1/Fu Zhuang 30 crosses, it is evidenced that UBC405-628 linked to Pm gene of Fu Zhuang 30 with a distance of 13 cM and a LOD value of 7.4. Even the genetic distance is 13 cM, before closer one be found, it can be used in molecular marker assisted selection of wheat breeding program.
