Screening of High Biomass and High Amino Acid Content Mutant Strain from Morchella conica

The isolation and regeneration of protoplasts from Morchella conica Pers. CGAC-9506 were studied.Some high biomass and high amino acid content mutant strains were isolated from M.conica CGAC-9506 protoplast treated by UV irradiation. The optimum condition of isolation protoplasts was that: 0.6 mol/L KCl phosphate buffer (pH 6.8), 4% snailase, 5% lywallzyme, 4% cellulase, hypha cultured 20 h , 30 ℃ digestion 3.5 h , and the yield of the protoplasts was 2.5×104 /mg. On the PSA (potato sucrose agar)medium containing 0.3 mol/L KCl+0.3mol/L inositol , the generation rate was 0.56% .The biomass and content of amino acids of M. conica CGAC - 950637,one of the mutant strains, were increased 7.3% and 38% respectively than that of the original strain.
