Inter-simple Sequence Repeat(ISSR) Analysis of Different Cultivars in Myrica rubra

Inter-simple sequence repeat (ISSR) was used to determine the genomic DNA variations and genetic relationships in Myrica rubra Sieb. et Zucc. Nine individuals including 7 cultivars and 2 clonal lines were used in ISSR analysis. A total of 30 ISSR primers were screened, of which 11 polymorphic and informative patterns were selected to determine the genetic diversity and genetic relationships. Total 116 DNA bands were amplified, 48 of which were polymorphic (41.4%). The average number of DNA bands amplified by each primer was 10.545. All cultivars tested were identified based on ISSR5 and ISSR16. DNA profiles based on ISSR markers had revealed potential diagnostic fingerprints for various individual accessions/cultivars. A tree diagram was constructed by using UPGMA algorithm. The Myrica rubra individuals including 7 cultivars and 2 clones were classified into 3 groups.
