Effects of Several Environmental Factors on the Activity of α-Galactosidase in Gracilaria Lemaneiformis (Rhodophyta)

Galactosidase (Galase) is one of the key enzymes for floridoside hydrolyzing in red algae. The effects of several environmental factors on Galase activity in Gracilaria lemaneiformis were investigated, such as light and dark cycle, salinity, temperature and nitrogen concentration of the culture media. It was shown that there was a strong diurnal variation of Galase activity: after 6 hours in the light period it reached the peak, in the following dark period it sustained at a low level and gradually changed. Galase activity was reduced after the algae were transferred to a hypersaline medium from a 33 salinity medium and raised after transferred to a hyposaline medium. Galase activity was dramatically affected by temperature; it showed higher activity at 30℃ or 15℃ than that at its usual grow temperatures. The effects of nitrogen concentration were different at different nitrogen levels.
