Purification and Properties of Cellulases from Stachybotrys Atra

After cultivating in liquid medium for 96h, three extracellular endo-β-1, 4-glucanase (EC 3.2.1.4) and a β-1,4-glucosidase (EC 3.2.1.21) from a Stachybotrys atra strain s607 was purified to electrophoretic homogeneity by Sephadex G100 column chromatography, DEAE-Sepharose fast flow ion-exchange chromatography, CM-Sepharose fast flow ion-exchange chromatography, Q-Sepharose fast flow ion-exchange chromatography and displacement chromategraphy . The enzymes were identified as EGⅠ? EGⅡ?EGⅢ and β Gase. The molecular weights of EGⅠ?EGⅡ?EGⅢ and β Gase were 78.3 kDa?58.1 kDa?72.4 kDa and 55.6 kDa respectively (by SDS-PAGE). The four components showed their maximum activities at their optimal pH 6.0?7.0?6.5?6.0 and optimal temperature 50℃?50℃?50℃?55℃ respectively. The three EGs can also hydrolyse xylan.
