Study on Reaction Catalyzed by Endoglucanase from Trichioderma Viride with Microcalorimetric Method

The endoglucanase of cellulase is separated from Trichioderma viride and purified.The influences of acidity,temperature and metal ions to the enzyme catalyzed reaction are studied with microcalorimetric method and the power-time curves of the enzyme catalyzed reaction are determined.According to the curves,the maximum reaction rate(Vmax) and the Michaelis constant(Km) of the reactions under different conditions are calculated by thermokinetics theory and contrast-progress method.The optimum acidity(pH=5.20) and optimum temperature(T=322.88 K) are obtained.The Vm-c equations describing the inhibitory action or activation of metal ions to the enzyme catalyzed reaction are obtained.
